流动相:乙腈(A)和0.1%磷酸溶液(B),梯度洗脱(0~40min,5%~15%A;40~80min,15%~28%A;80~100min,28%~28%A); 柱温为30℃; 流速为1.00ml/min; 对3批浮萍配方颗粒进行特征图谱测定,记录图谱。结果见图4。 4)分析特征图谱 浮萍配方颗粒特征图谱中呈现7个特征峰,与牡荆素参照物相应的峰为S峰,计算各特征峰与S峰的相对保留时间,其相对保留时间应在规定值的±8%范围之内。规定值为0.35(峰1)、0.47(峰2)、0.72(峰3)、0.91(峰4)、1.00(峰5,S峰)、1.07(峰6)、1.21(峰7)。 实施例5浮萍药材-饮片-提取物及配方颗粒对照图谱 采用中药色谱指纹图谱相似度评价系统(2012版)分别将实施例1中16批浮萍药材特征图谱合成一张药材对照图谱,实施例2中16批浮萍饮片特征图谱合成一张饮片对照图谱,实施例3中16浮萍标准汤剂特征图谱合成一张标准汤剂对照图谱,实施例4中3批浮萍配方颗粒特征图谱合成一张配方颗粒对照图谱。将以上浮萍药材、饮片、标准汤剂、配方颗粒对照图谱进行对比,结果见图5。 结果表明:饮片和标准汤剂、配方颗粒中的7个特征峰均能在药材中检出,说明这些成分都来源于药材,同时从对照图谱可知浮萍药材、浮萍饮片、浮萍标准汤剂、浮萍配方颗粒的化学成分相同,物质基础一致。 以下通过试验例具体说明本发明的有益效果: 试验例1特征图谱方法学 色谱条件与系统适用性试验以十八烷基硅烷键合硅胶为填充剂(柱长为250mm,内径为4.6mm,粒度为5μm)以乙腈为流动相A,以0.1%磷酸溶液为流动相B,按表1中的规定进行梯度洗脱,理论板数按牡荆素峰计算应不低于5000。 表1流动相梯度洗脱比例 1波长选择 在以上拟定的实验条件基础上,利用二极管阵列检测器分别对牡荆素、供试品溶液(批号1805034浮萍配方颗粒)进行全波段扫描,并分别提取供试品溶液在220nm、240nm、260nm、280nm、300nm、320nm、340nm波长下的色谱图。见图6-7。 结果表明,在检测波长为300nm时色谱峰信息量较大,色谱图基线更平稳,故检测波长确定为300nm。 2柱温考察 在以上拟定的实验条件基础上,取同一供试品溶液,分别对柱温为25℃、30℃、35℃时进行考察。见图8。 柱温考察结果表明,当柱温为30℃时,色谱图峰形均较为对称,分离度均好,故最终确定30℃作为浮萍特征图谱方法的柱温。 3流速考察 在以上拟定的实验条件基础上,取同一供试品溶液,分别对流速为0.8ml/min、1.0ml/min、1.2ml/min时进行了考察。见图9。 结果表明,流速在1.0ml/min时,色谱图峰形较好,分离度适中。故流速确定为1.0ml/min。 综上所述,浮萍特征图谱色谱条件与系统适应性试验确定为:以十八烷基硅烷键合硅胶为填充剂(柱长为250mm,内径为4.6mm,粒度为5μm);以乙腈为流动相A,以0.1%磷酸溶液为流动相B,梯度洗脱(0~40min,5%~15%A;40~80min,15%~28%A;80~100min,28%~28%A);柱温为30℃;检测波长为300nm。理论板数按牡荆素峰计算应不低于5000。 4参照物溶液的制备 取牡荆素对照品适量,精密称定,加50%甲醇制成每1ml含50μg的溶液,即得。 5供试品溶液的制备 5.1提取溶剂考察 取浮萍配方颗粒(批号1805034)0.2g,置具塞锥形瓶中,分别加入甲醇、75%甲醇、50%甲醇、水、乙醇、50%乙醇、75%乙醇各10ml,密塞,超声处理(功率600W,频率40kHz)30分钟,放冷,摇匀,滤过,取续滤液,即得。见图10。 结果表明,提取溶剂中50%甲醇作为提取溶剂时色谱峰信息量大,故供试品提取溶剂确定为50%甲醇。 5.2提取方式考察 取浮萍配方颗粒(批号1805034)0.2g,置具塞锥形瓶中,加入50%甲醇25ml,密塞,分别对供试品提取方法为回流、超声时进行考察,提取时间20min,放冷,摇匀,滤过,取续滤液,即得。见图11。 结果表明,对供试品分别进行超声提取与回流提取时效果一致。因超声提取操作更为简便,故供试品提取方法确定为超声提取。 5.3提取时间考察 取浮萍配方颗粒(批号1805034)0.2g,置具塞锥形瓶中,加入50%甲醇25ml,密塞,超声处理(功率600W,频率40kHz),分别对供试品提取时间为20分钟、30分钟、40分钟时进行考察,放冷,摇匀,滤过,取续滤液,即得。见图12。 结果表明,在提取时间为20分钟时,即可充分提取。故供试品提取时间确定为20分钟。 5.4溶剂加入量考察 取浮萍配方颗粒(批号1805034)0.2g,置具塞锥形瓶中,分别加入50%甲醇10ml、25ml、50ml,密塞,超声处理(功率600W,频率40kHz)20min,放冷,摇匀,滤过,取续滤液,即得。见图13。 结果表明,在溶剂加入量为25ml时,特征图谱色谱峰面积适中,基线更平。故供试品溶剂加入量确定为25ml。 5.5确定供试品制备方法:取浮萍制剂,研细,取0.2g,置具塞锥形瓶中,加入50%甲醇25ml,超声20分钟,放冷,摇匀,滤过,取续滤液,即得。 5.6特征图谱方法确定为 照高效液相色谱法(《中国药典》2015年版四部通则0512)测定。 色谱条件与系统适用性试验以十八烷基硅烷键合硅胶为填充剂(柱长为250mm,内径为4.6mm,粒度为5μm);以乙腈为流动相A,以0.1%磷酸溶液为流动相B,按下表中的规定进行梯度洗脱;柱温为30℃;检测波长为300nm。理论板数按牡荆素峰计算应不低于5000。 参照物溶液的制备取牡荆素对照品,精密称定,加50%甲醇制成每1ml含50μg的溶液,作为对照品参照物溶液。 供试品溶液的制备取浮萍药材、饮片粉末1g,置具塞锥形瓶中,加50%甲醇25ml,密塞,超声处理(功率600W,频率40kHz)20分钟,放冷,摇匀,滤过,取续滤液,即得。另取浮萍标准汤剂粉末、配方颗粒,研细,取0.2g,置具塞锥形瓶中,加入50%甲醇25ml,称定重量,超声处理(功率600W,频率40kHz)20分钟,放冷,摇匀,滤过,取续滤液,即得。 测定法分别精密吸取参照物溶液与供试品溶液各10μl,注入液相色谱仪,测定,即得。 试验例2特征图谱方法学考察 1色谱峰指认 供试品溶液的制备:按以上拟定的实验条件,制备浮萍配方颗粒供试品溶液。 参照物溶液的制备:取牡荆素对照品适量,精密称定,加50%甲醇制成每1ml含50μg的溶液,即得。 浮萍对照药材溶液的制备:取浮萍对照药材粉末1g,置具塞锥形瓶中,加入50%甲醇25ml,密塞,超声处理(功率600W,频率40kHz)20分钟,放冷,摇匀,滤过,取续滤液,即得。 阴性对照溶液的制备:按以上拟定的实验条件,制备缺浮萍配方颗粒阴性对照溶液。 对浮萍配方颗粒特征图谱峰进行定位。见图14-15。 2精密度试验 取浮萍配方颗粒供试品溶液,按拟定实验方法连续进样6次,每次10μl,计算各特征峰的相对保留时间及相对峰面积。见表2-3。 表2精密度考察-保留时间 表3精密度考察-峰面积 结果表明,该仪器精密度良好。 3重复性考察 精密称取浮萍配方颗粒6份,按拟定实验方法进行制备及测定。见表4-5。 表4重复性考察-相对保留时间比值 表5重复性考察-相对峰面积比值 结果表明,该方法重复性好。 4中间精密度考察 4.1不同仪器考察 在以上拟定的实验条件基础上,分别精密称取浮萍配方颗粒两份,制备供试品溶液,分别在安捷伦1260、岛津LC-20AD、Waters e2695型高效液相色谱仪上进行测定。见表6-7。 表6仪器耐用性考察-相对保留时间比值 表7仪器耐用性考察-相对峰面积比值 结果表明,用上述3种仪器对供试品进行检测时,各特征峰相对保留时间的RSD范围为0.0%-1.2%。 4.2不同人员和时间考察 在以上拟定的实验条件基础上,由不同人员(A、B)在不同时间(T1、T2)分别精密称取浮萍配方颗粒各两份,制备供试品,进行测定。见表8-9。 表8人员和时间考察-相对保留时间比值 表9人员和时间考察-相对峰面积比值 结果表明,由不同的人员在不同的时间对同一个样品进行测定,方法稳定性较好。 5耐用性考察 5.1色谱柱耐用性考察 在以上拟定的实验条件基础上,分别对色谱柱为Agilent TC-C18 5μm250×4.6mm、Diamonsil 5μm C18(2)250×4.6mm、Agilent Eclipse XDB-C18 5μm 4.6×250mm进行考察。见表10-11。 表10色谱柱耐用性考察-相对保留时间比值 表11色谱柱耐用性考察-相对峰面积比值 结果表明,用上述3种色谱柱对样品进行检测,特征峰相对保留时间的RSD在0.0%~9.8%。 5.2稳定性 在以上拟定的实验条件基础上,取同一供试品溶液,分别于0h,4h,7h,14h,20h,24h时测定。见表12-13。 表12稳定性考察-保留时间 表13稳定性考察-峰面积 结果表明,其相对应的特征峰保留时间的RSD为0.1%,峰面积RSD为0.1%-1.5%,样品溶液在24小时内稳定。 综上所述,各特征峰相对保留时间的RSD在以上各项考察中均符合要求,该方法良好。将上述8个特征峰纳入后续考察。 6特征峰的确定及对照图谱的建立 6.1相对保留时间规定值限度的制定 方法学各考察项目及验证结果汇总见表14。 表14方法学各项目结果RSD%汇总标准—相对保留时间 在色谱柱耐用性考察项中,峰1的RSD为9.8%,RSD值较大,因此不将峰1纳入特征峰范围内。除去峰1,峰2至峰8的RSD范围为0.0%-6.3%,故将各峰的相对保留时间规定值暂定为±8%。 最终规定:供试品特征图谱中应呈现7个特征峰,其中与牡荆素参照物相应的峰为S峰。 6.2 3批浮萍配方颗粒验证结果 采用拟定的方法对浮萍配方颗粒3批样品进行特征图谱的测定,计算相对保留时间、相对峰面积。见图16,表15-16。 采用中药色谱指纹图谱相似度评价系统(2012版)对3批浮萍配方颗粒进行合成,建立了浮萍配方颗粒特征图谱的对照图谱。见图17。 表15 3批浮萍配方颗粒相对保留时间 表16 3批浮萍配方颗粒相对峰面积 根据相对保留时间稳定及各批次样品均能检出且峰相对较高的原则,共选择了7个重复性较好的峰作为特征峰。计算各特征峰与S峰的相对保留时间,其相对保留时间应在规定值的±8%范围之内,规定值为0.35(峰1)、0.47(峰2)、0.72(峰3)、0.91(峰4)、1.00(峰5,S峰)、峰6)、1.21(峰7)。 通过特征图谱方法学考察,确认了该方法能检测浮萍配方颗粒中的7个特征峰;对浮萍药材、饮片、标准汤剂及浮萍配方颗粒均能检测7个稳定峰,说明浮萍药材、饮片、标准汤剂及配方颗粒的物质基础一致。该方法能用于药材-饮片-提取物及配方颗粒的质量检测,能准确反应浮萍药材-饮片-提取物及配方颗粒多成分的整体面貌。 Claims (10)1.一种浮萍及其制剂的特征图谱检测方法,其特征在于:它是采用高效液相色谱法检测,其操作步骤如下: 1)参照物溶液的制备:取牡荆素对照品,加50%甲醇溶解,即得;
